My rule of thumb is to at least "pinch an inch" of substrate along with the excrement; then, I will still check down to the bottom of the enclosure to see if anything's stuck to it, namely dried urine. If that is present, I clean about 2" to either side of the dried "matter", wipe with wet paper towels, then I use a disinfectant (strong bleach and water solution), and wipe with wet towels again to remove the strong chlorine smell. When all cleaning is done, I put in a spot of new substrate.
I replace all substrate at least once every six months, or more frequently if the snake is a very active "poop-er", like my False Water Cobra.
